Recombinant Alkali-tolerant Protein A (rat-PA)
Recombinant alkali-resistant protein A was modified on the basis of natural Staphylococcus aureus protein A. Compared with natural protein A, the binding site with Fab was removed, the specificity of recombinant alkali-resistant protein A and Fc was enhanced, and the protein stability and alkali resistance were improved. After the recombinant alkali-resistant protein A is bonded to the agarose gel, an affinity filler for antibody purification can be prepared, and antibodies of various mammalian subtypes or genetically engineered recombinant proteins containing antibody Fc fragments can be isolated and purified from ascites, serum, cell culture supernatant or cell extracts. A cysteine was added to the C- terminus of recombinant alkali-resistant protein A to enhance its directional covalent bonding with the gel.
