Ni-NTA and IMAC Affinity Resin
Ni-NTA NUPharose FF and IMAC NUPharose FF are immobilized metal affinity chromatography (IMAC) resins containing nitrilotriacetic acid (NTA) groups, which are used for affinity purification of his-tagged proteins.
IMAC NUPharose FF is free of metal ions (uncharged), allowing the user to charge it with the most appropriate metal ion for purification of a target protein, such as Ni2+, Co2+, Cu2+, Zn2+, etc. Ni-NTA NUPharose FF is precharged with Ni2+ and can be used directly.
Through the optimization of the ligand modification process, these two products have achieved a balance between affinity and specificity: the binding capacity of his-tagged proteins is greater than 40 mg/mL, and the sample purity can reach more than 90% after one-step purification. In addition to histidine, the resins can also form coordination bonds with tryptophan and cysteine residues, so these two resins can be used to purify proteins without tags but containing these amino acid residues, such as human apotransferrin.
